Cyclic Peptidomimetic Lead Compounds to Reduce Neurotoxicity and Associated Oxidative Stress in Alzheimer’s Disease
نویسندگان
چکیده
Cyclic peptidomimetic compounds have been investigated utilizing ab initio Hartree Fock molecular orbital calculations. Prospective use of these ionophores as curative drugs for Alzheimer's disease has been investigated by considering their metal toxicity removal efficiency. A (2,4,2,4) system that is, ((gly-et)-(ala-gly-gly-et))2 with CH2NH backbone is predicted to undergo low conformational reorganization in presence of Zn(2+) ion with moderate electrostatic stabilization of ion. Conformation of this system and molecular weight render it suitable to be a lead compound for metal toxicity removal drugs required in AD.

Review: Alzheimer’s Amyloid b-Peptide-Associated Free Radical Oxidative Stress and Neurotoxicity
s Alzheimer’s disease, the major dementing disorder of the elderly that affects over 4 million Americans, is related to amyloid b-peptide, the principal component of senile plaques in Alzheimer’s disease brain. Oxidative stress, manifested by protein oxidation and lipid peroxidation, among other alterations, is a characteristic of Alzheimer’s disease brain. Our laboratory united these two obser...
متن کاملpeptide-associated free radical oxidative stress
Amyloid b-peptide (Ab), the central constituent of senile plaques in Alzheimer’s disease (AD) brain, has been shown to be a source of free radical oxidative stress that may lead to neurodegeneration. In the current study Ab(1-40), found in AD brain, and the amyloid fragment Ab(25-35) were used in conjunction with electron paramagnetic resonance spin trapping techniques to demonstrate that these...
متن کاملCurcumin Ameliorates Sodium Valproate Induced Neurotoxicity through Suppressing Oxidative Stress and Preventing Mitochondrial Impairments
Background and purpose: Curcumin is a natural polyphenolic compound in turmeric (Curcuma longa). Curcumin has potent free radical scavenger and antioxidant properties that could significantly reduce oxidative damage. Oxidative stress and mitochondrial dysfunction contribute to valproate sodium induced tissue damage. This study investigated the protective effects of curcumin against valproate so...
متن کاملThe effect of flavonoids in the treatment of Alzheimer’s disease: review article
Alzheimer’s disease (AD) is the most prevalent age-related neurodegenerative disorder worldwide, and no cure or prevention has been found for it. Extracellular senile plaque and intracellular neurofibrillary tangles are two important histopathological hallmarks of AD, which are both harmful for the cell. Senile plaques are composed of amyloid beta and neurofibrillary tangles are formed by hyper...
متن کاملDiscovery of the first dual GSK3β inhibitor/Nrf2 inducer. A new multitarget therapeutic strategy for Alzheimer’s disease
The formation of neurofibrillary tangles (NFTs), oxidative stress and neuroinflammation have emerged as key targets for the treatment of Alzheimer's disease (AD), the most prevalent neurodegenerative disorder. These pathological hallmarks are closely related to the over-activity of the enzyme GSK3β and the downregulation of the defense pathway Nrf2-EpRE observed in AD patients. Herein, we repor...
